How Automated Nucleic Acid Clean-Up Works
The Microlab PuriFY transforms complex, error-prone workflows into a streamlined process. With minimal manual interaction you can achieve reliable, reproducible results. Here's how it works:
- Select Protocol: Choose your clean-up parameters using the simple touchscreen interface.
- Load Samples and Buffers: Transfer your bead/sample suspension into the Microlab PuriFY Strips, designed for precise handling and minimal waste. Load your preferred target labware and the required buffers.
- Load Checks: The Microlab PuriFY automatically checks liquid levels, ensuring sufficient buffer and waste capacities for the run. It detects the Microlab PuriFY Strips and all required labware to safely run the protocol.
- Run Protocol: With the press of a button, you can let the Microlab PuriFY take over the handling of each step with precision.
- Retrieve Purified Samples: Collect your clean, high-yield samples, ready for downstream applications.
A Deeper Look: MagPore Technology
MagPore, Magnetic Bead Technology
Simply transfer your magnetic bead/nucleic acid (NA) suspension into the Microlab PuriFY Strips and start your run. The Microlab PuriFY will manage the process starting with the initial bead/sample incubation.
- Bead Immobilization: Following the initial incubation, the magnetic racks are precisely positioned at the Microlab PuriFY Strip to effectively capture all magnetic beads in the suspension.
- Removal of Contaminants: After sealing the Microlab PuriFY Strip, pressure is applied into the individual vessels to transfer the supernatant through the vessel pore into the liquid waste container.
- Washing: The micro-dispense capillaries are positioned exactly above the vessels to dispense ethanolic wash buffer to the beads. Following the dispense, the wash buffer is removed into the liquid waste container using positive pressure while the magnetic beads remain captured.
- Resuspension: Following the final wash and subsequent air drying, elution buffer is dispensed into the vessels. Two magnetic racks move alternately up and down to allow the magnetic beads to repeatedly move through the solution ensuring high NA recovery rates.
- Nucleic Acid Elution: Following the final incubation step, magnetic beads are captured to allow the NA solution to be propelled through the pore using pressure. The target consumable is positioned below the Microlab PuriFY Strips in time to capture the purified NA.
