Target protein purification and enrichment prior are used prior to HPLC measurements after monoclonal antibody production. Affinity applications for IgG isolation, recombinant protein purification, anti-drug antibody assay, therapeutic drug monitoring provides a mild and reliable separation of interfering impurities. Moreover, the protein analysis workflows often require additional buffer exchange or removal of salts that can interfere with downstream processes and analytical measurements. Therefore, size exclusion chromatography is frequently used.

SizeX IMCStips® are miniaturized affinity and size exclusion pipette tips and are an effective method for protein analysis due to the mild separation conditions used to obtain high-resolution separations. Here, we demonstrate the reproducible and high-yield performance of Protein Affinity followed by SizeX IMCStips on a Hamilton Microlab STAR V liquid handling system by processing cell culture supernatant.

Hamilton Microlab STAR V allows high reproducibility and high yield of antibody recoveries after size exclusion chromatography using 8 individual pipetting channels and 96 Multiprobe Head. Hands-free purification and buffer exchange of up to two 96-well plates could be achieved in one run.
