Hamilton's PRP-Infinity HPLC columns simplify the separation of proteins, including difficult high molecular weight proteins. The non-porous support eliminates pore diffusion which can cause band broadening. This ensures narrow sharp peaks for even the largest proteins.
Extremely Fast Separations
The non-porous PRP-Infinity packing allows gradient elution of six proteins in 60 seconds. Application #255, #256 and #1 illustrate the effect of increasing flow rate on separation. As the flow rate is increased from 0.5 mL/min in Application #255 to 2.0 mL/min in Applications #1 and #256, the peak width narrows and the resolution improves
Enhanced Detectability
The short separation time (60 seconds) minimizes band broadening which keeps peaks narrow and detectability high. pH stability from 1 to 13 allows separation of proteins at or above their isoelectric point.
Separate High Molecular Weight Proteins
The non-porous PRP-Infinity support is free of the limitations a wide pore support has (band broadening and ghosting because of slow diffusion into the pores). This allows you to chromatograph large proteins without excessive band broadening or ghosting.
High Resolution
The column's resolving power is illustrated in Application #253, the separation of three Cytochrome c species (equine, bovine and canine). Cytochrome c is a 104 amino acid long protein. The difference between equine and canine is only six amino acids. It is remarkable that the protein can be partially resolved in less than five minutes. Application #254 shows the high pH separation of the same Cytochrome c species. While the resolution is not as good it is interesting to note the elution order change.
Micropreparative Use
PRP-Infinity can be used for isolation of micropreparative amounts of proteins that previously have been difficult to chromatograph (because of size). Since the support is non-porous, the sample capacity is approximately 20% that of a 300 Angstrom material.
