High-quality plasmid DNA is critical for downstream applications such as Sanger sequencing, cloning, and transfection. DNA extraction from cultures can be time-consuming when done manually. Automating the extraction process can greatly improve throughput, decrease cross contamination, and decrease user variability. The Microlab® STAR, as shown in Figure 1, is a flexible, automated robotic pipetting system that can be adapted to perform various vacuum or magnetic bead-based tasks. The STARline platform uses air displacement technology, which increases pipetting accuracy and eliminates sample contamination and dilution. Omega Bio-tek offers high-quality, fast, and economical plasmid DNA extraction kits that can be integrated into the Hamilton liquid handlers. Omega Bio-tek offers a complete line of high-throughput plasmid DNA extractions kits that incorporate either silica or magnetic bead-based technologies that can be tailored to the individual needs of the customer. Typically, Omega Bio-tek kits produce 10-15 μg high-copy plasmid DNA from a 1 mL overnight culture, although yields will vary according to plasmid copy number, bacterial strain, and growth conditions.
